Circuit properties and epileptogenesis in grey matter heterotopia

Abstract
Grey matter heterotopia (GMH), including subcortical band heterotopia (SBH) and periventricular nodular heterotopia (PVNH), are neurodevelopmental disorders associated with epilepsy. Using preclinical models, I investigated the mechanisms underlying epileptogenesis in SBH and compared the pathological properties of SBH and PVNH. In SBH, I identified age-dependent seizure progression and demonstrated that early, targeted modulation of neuronal excitability could influence epileptogenesis. Comparatively, I found that SBH and PVNH exhibit both shared and distinct physiological and morphological features, contributing to the formation of pro-epileptic networks. My findings highlight the heterogeneity of GMH subtypes and underscore the importance of early, tailored interventions to mitigate epilepsy in these disorders.
